To get a third level of Spiritus, yes.

In general, any vampire who already has a superior discipline can still gain an extra level of the discipline. It doesn't have any game-based benefit, though can provide greater resilience if you lose one of the levels somehow - such as here, if Denette stopped being Independent and lost the Field Training card as a result.

A similar ruling for Blooding by the Code.
