The quoted text for Gianna at the start of this thread is from 2014 and is thus now badly out of date.

The current text for Gianna reads

Unique wraith with 2 life. 1 strength, 1 bleed.
If another minion you control is in combat, Gianna can lock and burn herself to burn an equipment on the opposing minion, or lock during normal strike resolution to inflict 1 unpreventable damage on the opposing minion.

Since this specifies "normal strike resolution", Gianna cannot lock to inflict damage if the combat doesn't reach that stage (due to S:CE, minion going to torpor from first strike, or other effects).
